Industrial technology for obtaining fucoxanthin, a marine antitumor carotenoid

How to isolate pure crystalline fucoxanthin from the biomass of the diatom Cylindrotheca closterium. According to the authors, 1 g of crystals was obtained, with up to 99 % of the pigment in its main form.

Fucoxanthin is an orange pigment of marine algae. To study its properties, researchers need a pure pigment rather than a mixture of substances from the biomass. The marine diatom C. closterium can contain 2 % fucoxanthin by dry weight or more, so it is convenient as a raw material. The authors set out to work out a technology for obtaining crystalline fucoxanthin from its biomass.

The pigment was extracted from the biomass with alcohol, then water was added and the precipitate was separated in a centrifuge. The procedure was repeated until chlorophylls and other pigments were removed. The remaining orange-red fraction was recrystallized from a solvent mixture, and the crystals were washed and dried in a stream of argon. Purity and structure were confirmed by mass spectrometry and nuclear magnetic resonance.

According to the authors, after recrystallization from one solvent mixture the main (trans) and minor (cis) forms of the pigment were in a ratio of 95 : 4, and from another 99 : 0.5. The spectra matched those described in the literature. In total, 1 g of crystalline fucoxanthin was obtained.

The work describes a practical way to obtain pure pigment for further research.

Limitations. This is a conference abstract: without a full description of the methods and without statistics. The word "industrial" in the title is the authors' assessment; the scale of the process is not described in the abstract. The antitumor properties mentioned in the title were not studied in this work — the authors refer to the literature.
